제가 선정한 논문은 2025년 Nature Communications에 출판된 “Lung structural cells are altered by influenza virus leading to rapid immune protection following re-challenge”입니다.

Influenza A virus(IAV)에 감염되면 epithelial cell, fibroblast, endothelial cell과 같은 lung structural cell이 초기 antiviral response와 immune cell recruitment에 중요한 역할을 합니다. 그러나 일반적으로 immune memory는 T cell이나 B cell과 같은 immune cell을 중심으로 연구되어 왔으며, structural cell 자체에 감염 경험이 장기간 남아 이후 재감염에 영향을 줄 수 있는지는 충분히 규명되지 않았습니다. 따라서 IAV 감염 이후 lung structural cell에 장기적인 변화가 유지되는지 밝히는 것이 중요합니다.

본 연구에서 저자들은 IAV 감염 후 lung epithelial cell, fibroblast, blood endothelial cell에 장기간 유지되는 변화에 주목하였습니다. 특히 감염 이후에도 이들 structural cell에서 immune-related gene expression과 antigen processing and presentation 관련 기능이 유지되는지, 그리고 이러한 변화가 재감염 시 빠른 antiviral protection에 기여하는지 규명하고자 하였습니다.

먼저 IAV 감염 후 epithelial cell, fibroblast, blood endothelial cell을 분리하여 분석한 결과, 감염 40일 이후에도 세 종류의 structural cell 모두에서 transcriptional change가 유지되었으며, 특히 antigen processing and presentation pathway가 공통적으로 증가하였습니다. 또한 epithelial cell과 fibroblast에서는 transcription factor인 SpiB와 관련된 antigen presentation gene이 지속적으로 증가하였습니다. 이후 epithelial cell을 분석한 결과, ciliated cell, club cell, progenitor cell, ATII cell 등 여러 population에서 MHCII expression이 증가하였으며 일부 세포에서는 MHCI와 MHCII가 높은 수준으로 감염 30일 이후까지 유지되었습니다. 이를 통해 IAV 감염이 lung structural cell에 장기간의 immune-related imprint를 남긴다는 것을 확인하였습니다.

마지막으로 저자들은 primary infection에 WSN(H1N1), re-challenge에 X31(H3N2)을 사용하여 재감염 모델을 구축하였습니다. 이전에 IAV 감염을 경험한 mouse의 epithelial cell에서는 primary infection group보다 IAV nucleoprotein(NP)이 감소하였으며, T cell을 depletion한 경우에도 재감염 초기인 day 2에서 낮은 lung viral titer가 유지되었습니다. 또한 naive mouse와 이전에 IAV에 감염된 mouse에서 structural cell을 분리하여 in vitro에서 다시 IAV를 감염시킨 결과, 이전 감염을 경험한 epithelial cell에서는 IAV-NP positive cell이 감소하였습니다. 이를 통해 감염을 경험한 epithelial cell 자체가 재감염 시 virus를 더 빠르게 control하는 cell-intrinsic antiviral response를 나타낸다는 것을 확인하였습니다.

결론적으로 본 연구는 IAV 감염 이후 epithelial cell, fibroblast, endothelial cell과 같은 lung structural cell에 장기간의 transcriptional imprint가 유지되며, antigen processing and presentation 및 MHC expression과 관련된 변화가 지속된다는 것을 보여주었습니다. 특히 이전 감염을 경험한 lung epithelial cell은 재감염 시 T cell에 의존하지 않고 초기 virus를 더 빠르게 control할 수 있었으며, 동시에 T cell과 communication할 수 있는 기능도 유지하였습니다. 이를 통해 lung structural cell이 단순한 physical barrier를 넘어 이전 감염 경험을 기억하고 이후 감염에 더 빠르게 반응하는 immune memory-like property를 가질 수 있다는 새로운 concept을 제시하였습니다.

저는 일반적으로 immune memory가 T cell이나 B cell과 같은 immune cell의 특성으로 알려져 있는 것과 달리, lung epithelial cell과 같은 structural cell도 이전 감염의 흔적을 장기간 유지하고 재감염 시 더 빠른 antiviral response를 나타낼 수 있다는 점에 관심을 가지고 본 논문을 선정하였습니다.
